Dynamic modification of proteins with ubiquitin is an important regulator of the cellular response to DNA damage. Several recent studies have pointed to important, but so far relatively unexplored roles of ubiquitin in the timely removal and/or degradation of DNA repair factors and chromatin-associated proteins during and/or after DNA repair.
We have developed a mass spectrometric (MS) method to monitor site-specific changes in ubiquitylation after cellular perturbations and have employed this method to perform a first systematic investigation of protein ubiquitylation following DNA damage induced by UV light irradiation. We are currently employing MS-based proteomics as well as biochemistry and cell biology to study the roles of protein ubiquitylation in the regulation of protein homeostasis in the nucleus after genotoxic stress.
